I want to expand my shop but am seriously considering the cost and the process. When I view the design tab, to expand to 12x15 or 13x16 it costs 250,000 or 500,000 respectively!

What I want to know is, can I go directly to 13x16 pay the 500,000 and bypass the 12x15? Or am I required to expand to 12x15 then 13x16 thereby paying 750,000 overall?

no, u can't skip expansions.

Really? Are you saying that I must purchase the 250,000 expansion prior to the 500,000 expansion?

yes, or use gems (if that's what is used in Fashion Story) ;)

Expansions have to be done in order. You cannot skip an expansion option.
